Исследования, направленные на создание и верификацию моделей диабета первого типа (СД1), играют важную роль в разработке новых методов лечения этого заболевания. Модели крупных животных могут сократить разрыв между фундаментальными исследованиями и клиническими испытаниями новых технологий для лечения дефицита инсулина. На данный момент наиболее востребованной моделью является стрептозотоцин-индуцированный диабет. Однако разные виды и породы животных по-разному реагируют на стрептозотоцин (СТЗ). Таким образом, несмотря на то, что уже существуют модели на разных породах свиней, верификация модели стрептозотоцин-индуцированного диабета на свиньях Wiesenau является важной задачей, так как эта порода еще не была использована, а также она является одной из наиболее доступных пород лабораторных свиней в России.</p></sec><sec><title>ЦЕЛЬ</title><p>ЦЕЛЬ. Воспроизведение модели стрептозотоцин-индуцированного диабета на свиньях Wiesenau с коррекцией гипергликемии инсулинотерапией и оценка ее стабильности и соответствия экспериментальной модели СД1.</p></sec><sec><title>МАТЕРИАЛЫ И МЕТОДЫ</title><p>МАТЕРИАЛЫ И МЕТОДЫ. Исследование было выполнено на 6 свиньях Wiesenau. Диабет индуцировался внутривенным введением СТЗ (150 мг/кг). Концентрацию глюкозы в крови определяли глюкометром, а также использовали систему непрерывного мониторинга глюкозы. Содержание инсулина в крови определяли иммуноферментным анализом (Cloud-clone corp). Успешность индукции диабета была подтверждена тестом толерантности к глюкозе. Через 7 дней после введения СТЗ начинали инсулинотерапию, которая продолжалась в течение трех месяцев. Через 3 месяца после индукции проводилось гистологическое и иммуногистохимическое исследование состояния островков поджелудочной железы.</p></sec><sec><title>РЕЗУЛЬТАТЫ</title><p>РЕЗУЛЬТАТЫ. Исследования на свиньях породы Wiesenau показали, что доза 150 мг/кг СТЗ достаточно безопасна и эффективно вызывает СД в 67% случаев. Был подобран оптимальный индивидуальный протокол инсулинотерапии, который успешно снижает уровень глюкозы в крови до нормальных значений. Эта модель стрептозотоцин-индуцированного диабета у свиней породы Wiesenau стабильна в течение 90 дней и может быть использована для исследования новых антидиабетических средств.</p></sec><sec><title>ЗАКЛЮЧЕНИЕ</title><p>ЗАКЛЮЧЕНИЕ. Сформирована модель стрептозотоцин-индуцированного диабета у свиней Wiesenau. Подтверждена стабильность модели в условиях снятия глюкозотоксичности инсулинотерапией.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>BACKGROUND</title><p>BACKGROUND: Research aimed at creating and validating models of diabetes plays an important role in the development of new treatments for this disease. Large animal models may bridge the gap between basic research and clinical trials of new technologies for the treatment of insulin deficiency. At the moment, the most popular model is streptozotocin-induced diabetes. However, different species and breeds of animals respond differently to streptozotocin. Thus, although models already exist in different breeds of pigs, verification of the Wiesenau pig model of streptozotocin-induced diabetes is an important task, since this breed has not yet been used, and it is also one of the most accessible breeds of laboratory pigs in the world. Russia.</p></sec><sec><title>AIM</title><p>AIM: To reproduce the model of streptozotocin-induced diabetes in Wiesenau pigs with correction of hyperglycemia by insulin therapy and evaluate its stability and compliance with the experimental model of type 1 diabetes.</p></sec><sec><title>MATERIALS AND METHODS</title><p>MATERIALS AND METHODS: The study was performed on 6 Wiesenau pigs. Diabetes was induced by intravenous administration of streptozotocin (150 mg/kg). Blood glucose concentration was determined with a glucometer and a continuous glucose monitoring system was used. The content of insulin in the blood was determined by enzyme-linked immunosorbent assay (Cloud-clone corp). The success of diabetes induction was confirmed by a glucose tolerance test. Seven days after the injection of STZ, insulin therapy was initiated and continued for three months. Insulin therapy was carried out for three months. 3 months after induction, a histological and immunohistochemical study of the condition of the pancreatic islets was carried out.</p></sec><sec><title>RESULTS</title><p>RESULTS: A study in Wiesenau pigs showed that a dose of 150 mg/kg streptozotocin was safe and effective in inducing diabetes in 67% of cases. An optimal individual insulin therapy protocol was selected, which successfully reduces blood glucose levels to normal values. This model of streptozotocin-induced diabetes in Wiesenau pigs is stable for 90 days and can be used to study new antidiabetic agents.</p></sec><sec><title>CONCLUSION</title><p>CONCLUSION: A model of STZ-induced diabetes has been established in Wiesenau pigs.
